All starches followed Type-III sorption isotherm with open. Thus, added lubrication fraction and adopted multivariate methodology should be selected carefully during the calibration and validation stages as it may have a significant impact on the predictability of the developed models.īreaking force multivariate analysis near-infrared spectroscopy partial least square regression principle component regression tableting.Ĭopyright © 2017 American Pharmacists Association®. grades (SPRESS B818, SPRESS B820, and SPRESS B825) were studied for. PURE-COTE and Instant PURE-COTE modified starches are for aqueous film coating and other. In addition, PLS calibration models' predictability was different from PCR calibration models. Spress B820 is a versatile, direct-compression tableting excipient. Predictability of PLS comprehensive (0.25%-1% w/w MgSt) model was significantly different from individual 0.25%, 0.5%, and 1.0% w/w MgSt PLS models. Expected qualitative correlations between MgSt blending time and TBF and a nonlinear relationship between MgSt fraction and TBF were observed. The predictability of calibration models was validated with independent data set. An oral dosage form can be readily prepared by dry-mixing and directly compressing.

Until now, high amylose starch (more than 70 amylose) has been used as the reaction substrate in most cases of CMS preparation. A qualitative principal component analysis and quantitative principal component regression (PCR) and partial least square (PLS) regression relationship between lubricant concentration, blending time, compression force, preprocessed NIR spectra, and measured TBF was calculated with calibration data set. Carboxymethyl starch CMS is a pH-sensitive tablet excipient that modulates drug release according to the physiological pH values. Tablets were scanned in reflection mode on a benchtop NIRS. In addition, we used a commercially available pregelatinized corn starch called Spress B820 (lot S0615476 GPC, Muscatine, IA, USA), as well as the product Avicel PH 101 (lot 11363 Fluka, Cork, Ireland). Starch 1500 Partially Pregelatinized Maize Starch Starch 1500 is a proven and trusted unique excipient, manufactured exclusively for the global pharmaceutical and nutritional industries.

Tablets of lubricated SPRESS ® B820 were prepared by varying lubrication and compression conditions using 2 4 full factorial design. This study used a local sago starch (food grade Nee Seng Ngeng & Sons, Sago Industries Sdn Bhd) and pregelatinized sago starches. Spress B820 and pregelatinised sago starch showed angles of repose of (30.23 0.46) and (30.37 0.23), respectively, and were rated as having excellent flow, and Avicel PH 101, with an angle of repose of (41.87 0.51), was rated as passable.

The predictability of multivariate calibration models, calculated with offline near-infrared spectroscopy (NIRS), assessing impact of magnesium stearate (MgSt) fraction, blending time, and compression force on the tablet breaking force (TBF) of SPRESS ® B820 was statistically compared.
